Cytogenetics of Switchgrass
We have analyzed cells in prophase stage from the actively growing root tips of switchgrass through generating chromosome squashes that were imaged with light microscopy. Preperation of Root Tips for Microscopy.
Karyotyping Switchgrass with ImageJ

Frame 1 - Raw image is uploaded into the program ImageJ.
Frame 2 - A threshold image is generated
Frame 3 - Juxtaposed chromosomes are manually seperated.
Frame 4 - Run “Analyze Particles” plug-in and send results to ROI manager.
Frame 5 - Use the ROI manager to make adjustments, measurements, and outlines.
Frame 6 - Generate overlay image of chromosome counts over raw image as final check.
Frame 7 - Straighten the chromosomes from the raw image
Frame 8 - Combine each straigtened chromosome into a karyotype
